Overview of PRLR molecular targets
Prolactin receptor (PRLR) is a cytokine receptor encoded by a gene located on chromosome 5p13-14, which can interact with prolactin as a transmembrane receptor. PRLR includes an extracellular region that can bind to prolactin, a transmembrane region, and a cytoplasmic region. PRLR is widely expressed in the breast, placenta, kidney, liver, and pancreas. In addition to prolactin, it can also be activated by growth hormone (GH) and human placental lactogen (hPL). Studies have shown that abnormalities in PRLR are closely related to the incidence and prognosis of certain tumors, especially breast cancer. Specific binding to PRLR can stimulate the growth of breast cancer cells, and some antagonists that can bind to PRLR but do not form dimers can inhibit the growth of breast cancer cells. There are also several isomers of PRLR, among which soluble PRLR cannot transduce prolactin signals.
