  • Overview of GDNF molecular targets


    Glial cell line-derived neurotrophic factor (GDNF) is a neurotrophic factor that promotes the survival of different neuronal subpopulations at different stages of development of the central and peripheral nervous systems. The functional human GDNF ligand is a disulfide-linked homodimer consisting of two 15kDa polypeptide chains. GDNF is produced by supporting cells, type 1 astrocytes, Schwann cells, neurons, pinealocytes, and skeletal muscle cells. It is a secretory protein that promotes the survival of motor neurons, midbrain dopaminergic neurons, Purkinje cells, and sympathetic neurons. GDNF is 100 times more efficient than neurotrophins in the survival of spinal motor neurons. In a mouse model of Parkinson's disease, GDNF was found to improve conditions such as bradykinesia, rigidity, and postural instability.
