Overview of FGF19 molecular targets
Fibroblast growth factor 19 (FGF-19) is encoded by the FGF-19 gene in humans. FGF-19 regulates bile acid synthesis and inhibits CYP7A1 in the liver through the FGFR4/Klotho-β receptor complex. In mouse models, FGF-19 also has metabolic effects, affecting glucose and lipid metabolism. FGF-19 is reduced in patients with chronic diarrhea due to bile acid malabsorption. FGF-19 is also present in the liver of patients with cholestasis. FGF-19 expression is reduced in patients with metabolic syndrome, non-alcoholic fatty liver disease, and insulin resistance. However, FGF-19 is increased to normal levels in obese patients who undergo Roux-en-Y gastric bypass surgery.
