  • Review of IGFBP1 molecular targets

Insulin-like growth factor-binding protein 1 (IGFBP-1), also known as placental protein 12 (PP12), is a protein encoded by the IGFBP1 gene. It binds to insulin-like growth factors (IGFs) I and II and circulates in the plasma. Such binding prolongs the half-life of IGFs and alters their interaction with cell surface receptors. IGFBPs can regulate the biological activity of IGF proteins. Post-transcriptional modifications of IGFBPs (such as glycosylation, phosphorylation, and proteolysis) can change the affinity of the binding protein to IGF. High concentrations of IGFBP-1 may be a marker of a catabolic state, and studies have shown that women with high concentrations of IGFBP-1 have less muscle mass. A higher IGFBP-1/IGF-1 ratio in the serum of patients with nasopharyngeal carcinoma is significantly associated with a poor prognosis. It has been reported that local IGFBP1 levels have an effect on the formation of coronary artery plaques.
