Overview of TIMP1 molecular targets
Matrix metalloproteinase inhibitor-1 (TIMP-1) is a glycoprotein expressed in various tissues of organisms and is a member of the TIMP family. TIMP-1 is a natural inhibitor of matrix metalloproteinases (MMPs), a group of peptidases involved in the degradation of the extracellular matrix. In addition to inhibiting most known MMPs, TIMP-1 can also promote cell proliferation in various cell types and may also have anti-apoptotic effects. Transcription of this gene can be strongly induced in response to many cytokines and hormones. Studies have shown that TIMP-1 gene inactivation in human females is polymorphic. ACTH, a nutrient hormone for adrenal cortical cells, induces the expression of TIMP-1, and increased TIMP expression is associated with decreased collagenase activity. In addition, increased TIMP-1 expression is associated with a poor prognosis in a variety of tumors such as laryngeal cancer or melanoma.
