Overview of PDGFRB molecular targets
Platelet-derived growth factor receptor β (PDGFRB), also known as CD140b, and the structural homologous protein PDGFRA (CD140a) are members of the class III subfamily of receptor tyrosine kinases (RTKs), which also includes receptors for M-CSF, SCF, and Flt-3 ligand. All class III RTKs contain five immunoglobulin-like domains in the extracellular region and a kinase domain in the intracellular region. PDGF is an important serum mitogen that can form homo- or heterodimeric proteins by linking the PDGF-A chain and PDGF-B chain through disulfide bonds. PDGFRA and PDGFRB bind to PDGF-AA, PDGF-BB, and PDGF-AB isomers with different affinities, with PDGFRA binding to all three PDGF isomers with high affinity, while PDGFRB can only bind to PDGF-BB with high affinity. PDGF binding can induce receptor homo- and heterodimerization and signal transduction. PDGFRA and PDGFRB are independently regulated in a variety of cell types. Recombinant soluble PDGFRB binds PDGF with high affinity and is a potent PDGF antagonist.
