  • Overview of ZAP70 molecular targets


    TCR-associated protein-70, expressed in T cells and NK cells, is required for TCR signaling and development. ZAP-70 interacts with the TCR complex by binding to ITAMs present in the invariant subunits of the TCR complex. Upon activation, ZAP-70 is phosphorylated on several tyrosine residues by two mechanisms: autophosphorylation and trans-phosphorylation by the Src family tyrosine kinases Lck1-3. Tyrosine phosphorylation of ZAP-70 is associated with its increased kinase activity and triggers downstream signaling events. Although ZAP-70 is usually expressed in T cells and NK cells, several recent studies have also shown that ZAP-70 positive expression is highly correlated with the expression of mutant IgVH in B chronic lymphocytic leukemia (CCL). In conclusion, the expression of ZAP-70 can be measured by intracellular flow cytometry and can serve as a prognostic marker for B-CLL.
