Dickkopf-related protein 1 (Dkk-1) is a secreted protein containing two cysteine-rich domains that participates in embryonic development through its inhibition of the Wnt signaling pathway. All homozygous Dkk-1 knockout mice die at birth due to structural defects in the skull and neural crest, such as the failure to develop the eyes, olfactory placodes, frontonasal region, and mandible, as well as incomplete development of the forebrain and midbrain and fusion of the toes of the forelimbs. Increased expression of Dkk-1 in the bone marrow, plasma, and peripheral blood is associated with osteolytic lesions in patients with multiple myeloma.
