Overview of P4HB molecular targets
Protein disulfide isomerase, also known as proline 4-hydroxylase beta subunit (P4HB), cellular thyroid hormone binding protein (p55) and glutathione-insulin transhydrogenase, is a peripheral membrane protein belonging to the protein disulfide isomerase family. It contains two thioredoxin domains and catalyzes the formation, cleavage and rearrangement of disulfide bonds. P4HB is localized near the CD4-rich region on the surface of lymphocytes. In the melanosome fraction, P4HB can reduce and activate the fusion properties of HIV-1 gp120 surface protein, thereby allowing HIV-1 to enter cells. P4HB acts as a chaperone to inhibit the aggregation of misfolded proteins. P4HB may participate in the structural modification of TG precursors in hormone biosynthesis with other molecular chaperones.
