NuRD Complex

The transcriptional repressor NuRD (nucleosome remodeling and histone deacetylase) complex is composed of multiple subunits, which include CHD3 (also known as Mi2-α), CHD4 (also known as Mi2-β), HDAC1, HDAC2, RBAP48, RBAP46, MTA1, MTA2, MTA3, and MBD3. Both CHD3 and CHD4 contain two PHD (plant homeodomain) zinc finger domains that bind directly to HDAC1 and HDAC2. HDAC1 and HDAC2 are highly homologous and are involved in histone deacetylation, chromatin remodeling and transcriptional repression, and are also found together in the mSin3A and MeCP1 complexes. RBAP46 and RBAP48 bind to the histone fold region of histone H4 and are believed to target the NuRD complex to its histone substrates. MBD3, a member of the methyl-CpG-binding domain protein family, and the metastasis associated genes MTA1, MTA2, and MTA3 were all found to be integral components of the NuRD complex.
