  • Overview of KDR molecular targets


    Vascular endothelial growth factor receptor 2 (VEGFR2), also known as kinase insert domain receptor (KDR), is a member of the class III subfamily of tyrosine kinase receptors (RTKs). Compared with VEGFR1, VEGFR2 is more widely expressed in endothelial cells derived from blood vessels. VEGFR2 binds to VEGF-A (VEGF121, VEGF165, VEGF189 and VEGF206 splice variants), VEGF-C and VEGF-D. Soluble VEGFR1 and VEGFR2 differ significantly in blocking VEGF-induced cell proliferation and migration. In contrast to soluble VEGFR1, soluble VEGFR2 is unable to compete with VEGF and binds to endothelial cells expressing VEGFR1 and VEGFR2. Soluble VEGFR2 can only partially inhibit cell migration, while soluble VEGFR1 almost completely blocks VEGF-induced cell proliferation and migration. The binding of VEGF to soluble VEGFR2 but not to VEGFR1 is also dependent on heparin. The VEGF/VEGFR2 signaling pathway plays an important role in tumorigenesis and other diseases with pathological angiogenesis. Inactivation of functional VEGFR2 using blocking antibodies can disrupt angiogenesis and prevent the spread of tumor cells.
