Function
Amphiregulin (AREG), encoded by the AREG gene on chromosome 4q13, is an epidermal growth factor (EGF)-family ligand of the ErbB/HER receptor tyrosine kinases. It is synthesized as a transmembrane pro-AREG precursor tethered to the plasma membrane; the extracellular EGF-like domain is then proteolytically shed chiefly by ADAM17 (TACE), and to a lesser extent ADAM10, releasing mature soluble AREG. Soluble AREG binds and activates EGFR (ERBB1) and also ERBB4, triggering receptor dimerization and autophosphorylation that propagate signals through the Ras/MAPK-ERK and PI3K-Akt pathways, as well as mTOR, leading to cell proliferation, survival, migration, and differentiation. AREG is a critical autocrine and paracrine growth factor for epithelial cells, keratinocytes, astrocytes, Schwann cells, and fibroblasts, and it plays central roles in wound repair and tissue regeneration, mammary-gland development, lung and gut mucosal healing, and resolution of inflammation, including repair mediated by activated regulatory T cells and type-2 innate lymphoid cells; it is also implicated in tissue fibrosis and cancer. AREG exists both as a membrane-anchored precursor on the producing cell and as a soluble shed growth factor acting in the extracellular space in an autocrine/paracrine manner.
