Function
Apolipoprotein M (ApoM), encoded by the APOM gene in humans, is a 25-kDa lipocalin-family apolipoprotein. It is produced mainly by hepatocytes and, to a lesser extent, by kidney proximal tubular cells, and in plasma it is associated predominantly with HDL (more than 90%) and to a minor extent with LDL, VLDL, and chylomicrons. Uniquely among apolipoproteins, ApoM retains its N-terminal signal peptide as a hydrophobic anchor that tethers it to the lipoprotein surface rather than being cleaved. Its best-characterized function is serving as the principal plasma carrier of the bioactive lysophospholipid sphingosine-1-phosphate (S1P): ApoM binds S1P inside its lipocalin calyx and delivers it to S1P receptors (S1PR1 to S1PR5), which are G-protein-coupled receptors. The ApoM-S1P axis signals through endothelial S1PR1/Gi to maintain the vascular endothelial barrier, suppress endothelial inflammation, regulate endothelial nitric-oxide production and vascular tone, and influence lymphocyte trafficking, angiogenesis, and wound healing. ApoM also contributes to HDL biogenesis and normal HDL size distribution. ApoM is a secreted plasma protein that remains loosely lipoprotein-associated, and the ApoM-S1P complex circulates on HDL.
