Function
Chondroitin sulfate proteoglycan 4 (CSPG4; also known as NG2 in rodents, HMW-MAA for high-molecular-weight melanoma-associated antigen, or MCSP) is a large transmembrane chondroitin-sulfate-bearing proteoglycan encoded by the human CSPG4 gene on chromosome 15. It is a single-pass type I membrane core protein with a huge extracellular domain decorated with chondroitin sulfate glycosaminoglycan chains, a short transmembrane segment, and a cytoplasmic C-terminal tail; it is expressed on the plasma membrane of melanoma cells, oligodendrocyte precursor cells, vascular pericytes and certain activated macrophages. Functionally CSPG4 acts as a co-receptor and signaling scaffold: its extracellular region binds and presents growth factors such as PDGF and FGF to their receptor tyrosine kinases and interacts with integrins and collagens (notably type VI), while its cytoplasmic tail couples to FAK, ERK1/2, CDC42/ACK1/BCAR1 and Rho-GTPase pathways that regulate cell adhesion, spreading, migration, polarization, proliferation and survival. On melanoma it promotes invasion, matrix remodeling and angiogenesis and is a major tumor-associated antigen. Subcellularly, CSPG4 is an integral cell-surface membrane proteoglycan.
