Function
Death receptor 3 (DR3), also known as tumor necrosis factor receptor superfamily member 25 (TNFRSF25) or TRAMP, is a type I transmembrane death receptor of the TNF receptor superfamily encoded by the human TNFRSF25 gene. It is preferentially expressed on activated and antigen-experienced T lymphocytes, FoxP3+ regulatory T cells, and some innate lymphoid cells. DR3 contains extracellular cysteine-rich domains and a cytoplasmic death domain, and its only known ligand is TNF-like cytokine 1A (TL1A/TNFSF15), a transmembrane and soluble cytokine rapidly upregulated on antigen-presenting cells and endothelial cells following TLR or Fc receptor activation. TL1A binding trimerizes DR3, which engages the adaptor TRADD and the kinase RIPK1, recruiting TRAF2 and cIAP1/2 to activate both canonical NF-kappaB and MAPK (ERK, JNK, p38) pathways that promote T cell proliferation, survival, and differentiation, particularly Th1, Th2, and Th17 responses. Under some circumstances, the DR3 death domain can also recruit FADD and caspase-8 to trigger apoptosis. DR3-TL1A signaling is central to mucosal immunity, intestinal inflammation, and regulatory T cell homeostasis. DR3 localizes to the plasma membrane of activated lymphocytes; TL1A exists in both membrane-bound and soluble forms.
