Function
Cadherin 1 (CDH1), commonly known as E-cadherin or epithelial cadherin (CD324), is encoded by the human CDH1 gene on chromosome 16q22.1. It is a 120 kDa type I transmembrane glycoprotein and the principal component of epithelial adherens junctions. Its extracellular five cadherin repeats mediate calcium-dependent homophilic adhesion between neighboring epithelial cells, while its cytoplasmic tail binds beta-catenin (or gamma-catenin/JUP), which in turn recruits alpha-catenin to anchor the complex to the actin cytoskeleton; p120-catenin binds the juxtamembrane domain to stabilize the junction and regulate actin dynamics. E-cadherin maintains epithelial polarity and tissue integrity, acts as a potent suppressor of epithelial-mesenchymal transition (EMT) and invasion, and is a well-established tumor suppressor; its somatic or epigenetic loss is a hallmark of diffuse gastric carcinoma and invasive lobular breast carcinoma.
