Function
Vitamin D-binding protein (DBP), also known as Gc-globulin (group-specific component) and encoded in humans by the GC gene, is a multifunctional plasma alpha2-globulin of the albumin gene family (alongside albumin, alpha-fetoprotein, and afamin). It is synthesized predominantly by hepatocytes and secreted into the blood, where it is the most abundant carrier of vitamin D metabolites; it is also present in ascitic and cerebrospinal fluid. It binds ergocalciferol (vitamin D2), cholecalciferol (vitamin D3), 25-hydroxycholecalciferol (calcifediol, 25-OH-D), and 1,25-dihydroxyvitamin D (calcitriol) at a single sterol-binding site, transporting them between skin, liver, kidney, and target tissues and modulating their bioavailability. Additional functions include scavenging extracellular G-actin released from damaged cells (preventing actin polymerization and intravascular coagulation), enhancing the neutrophil chemotactic activity of the complement fragment C5a, and serving as the precursor of Gc-protein-derived macrophage-activating factor (GcMAF). It is a secreted circulating plasma protein.
