Function
Interleukin-22 (IL-22), encoded by the human IL22 gene on chromosome 12q15, is a secreted alpha-helical cytokine of the IL-10 family. It is produced by leukocytes, chiefly Th17 and Th22 CD4+ T cells, gamma-delta T cells, NKT cells, and group 3 innate lymphoid cells (ILC3s), and is released systemically but acts predominantly on non-hematopoietic tissue cells. IL-22 signals through a heterodimeric receptor of IL-22R1 (IL-22RA1) and the shared IL-10R2 (IL-10RB) chain; IL-22R1 expression is largely restricted to epithelial cells, keratinocytes, hepatocytes, and stromal cells, explaining its tissue-targeted action. Receptor engagement activates JAK1 and Tyk2, leading to phosphorylation of STAT3 (and STAT1/STAT5), as well as PI3K/AKT and MAPK pathways, which induce epithelial cell survival and proliferation and production of antimicrobial peptides such as S100 proteins, Reg3beta/gamma, and beta-defensins. A soluble inhibitor, IL-22 binding protein (IL-22BP), neutralizes the cytokine. IL-22 is a secreted cytokine that bridges immune cells with barrier epithelia.
