Nectin-2 (nectin cell adhesion molecule 2; gene symbol PVRL2, also called CD112 or PRR2, formerly herpesvirus entry mediator B/HveB) is a member of the nectin family of immunoglobulin-like cell adhesion molecules, encoded in humans by the PVRL2 gene on chromosome 19q13. It is a single-pass type I transmembrane glycoprotein containing three extracellular Ig-like domains (one V-type and two C2-type), a transmembrane helix, and a cytoplasmic tail that binds the actin-binding adaptor afadin. Nectin-2 localizes primarily at adherens junctions (zonula adherens) of epithelial and endothelial cells, where it initiates calcium-independent cell-cell contact through trans-homophilic (Nectin-2/Nectin-2) and trans-heterophilic (Nectin-2/Nectin-3, and Nectin-2/Nectin-1) interactions, cooperating with E-cadherin-catenin complexes to organize apical junctional architecture and tight junctions. In the immune system, nectin-2/CD112 on target cells serves as a cell-surface ligand for the activating receptor DNAM-1/CD226 on NK cells and CD8+ T lymphocytes, and is also engaged competitively by the inhibitory receptors TIGIT, CD96, and CD112R (PVRIG), thereby setting a balance between NK/T-cell activation and inhibition. Nectin-2 also acts as an entry receptor for certain strains of herpes simplex virus and pseudorabies virus, facilitating viral cell-cell spread. Subcellularly it is a plasma membrane cell-cell junctional adhesion molecule; soluble isoforms generated by alternative splicing or shedding can be found extracellularly.
