Function
Tumor necrosis factor receptor superfamily member 14 (TNFRSF14/HVEM/CD270, herpesvirus entry mediator), encoded by the TNFRSF14 gene in humans, is a bidirectional immune co-signaling receptor. It is a single-pass type I transmembrane protein expressed on the plasma membrane of T cells, B cells, NK cells, myeloid cells, and endothelial cells. HVEM engages four structurally distinct ligands: the TNF-superfamily cytokines LIGHT (TNFSF14) and homotrimeric LT-alpha deliver co-stimulatory signals that activate NF-kB and promote T-cell proliferation and survival, whereas the Ig-superfamily molecules BTLA and CD160 deliver inhibitory signals that restrain T-cell activation. HVEM also serves as the cell-entry receptor for herpes simplex virus via its envelope glycoprotein D. By toggling between stimulatory and inhibitory ligand interactions, HVEM acts as a molecular switch regulating T-cell activation, mucosal immunity, and tolerance.
