Function
Thrombospondin-1 (TSP-1), encoded by the human THBS1 gene, is a ~450 kDa trimeric matricellular glycoprotein. Each subunit contains an N-terminal heparin-binding domain, a procollagen homology domain, three properdin-like type I repeats (TSRs), four EGF-like repeats, a high-affinity calcium-binding type III repeat region, and a C-terminal cell-binding domain. TSP-1 is secreted by platelets (stored in alpha-granules and released upon activation), endothelial cells, smooth muscle cells, fibroblasts, and macrophages, and is incorporated into the extracellular matrix. TSP-1 engages multiple cell-surface receptors: CD36 on microvascular endothelial cells (triggering Fyn kinase, p38 MAPK, and caspase-mediated endothelial apoptosis that underlies its potent anti-angiogenic activity); CD47 (integrin-associated protein), through which it inhibits nitric oxide/cGMP signaling in vascular cells and blunts ischemic responses; integrins alphavbeta1/alphaIIbbeta3; and low-density lipoprotein receptor-related protein 1 (LRP1). TSP-1 also binds and activates latent transforming growth factor-beta (TGF-beta), regulates platelet aggregation, wound healing, synaptogenesis, and tumor growth, and is a major endogenous inhibitor of angiogenesis.
