Function
Vascular endothelial growth factor receptor 1 (VEGFR1/FLT1), encoded by the human FLT1 gene, is a class III receptor tyrosine kinase. It is a type I transmembrane protein with seven extracellular immunoglobulin-like domains, a single transmembrane segment, and an intracellular tyrosine kinase domain, expressed on vascular endothelial cells, placental trophoblasts, monocytes/macrophages, and vascular smooth muscle cells. FLT1 binds VEGF-A, VEGF-B, and placental growth factor (PlGF) with high affinity; its intrinsic kinase activity is relatively weak, and it functions largely as a decoy or modulatory receptor that sequesters VEGF-A away from VEGFR2 and fine-tunes angiogenic signaling. Alternatively spliced soluble isoforms (sVEGFR1/sFlt1) lacking the transmembrane and kinase domains are constitutively secreted by endothelial cells and act as circulating VEGF/PlGF scavengers; excess sFlt1 in preeclampsia causes maternal endothelial dysfunction. Membrane-bound FLT1 also mediates monocyte and macrophage chemotaxis toward VEGF gradients. Subcellularly, FLT1 exists as a plasma membrane-bound full-length receptor and as a soluble secreted circulating form.
