Function
von Willebrand factor (VWF), encoded by the human VWF gene, is a large multimeric adhesive plasma glycoprotein essential for hemostasis. It is synthesized in endothelial cells and megakaryocytes: in endothelial cells it is stored in Weibel-Palade bodies and either constitutively secreted or released upon stimulation; in megakaryocytes it is packaged into platelet alpha-granules. VWF undergoes extensive post-translational processing including dimerization, multimerization, and glycosylation, with ultra-large multimers being the most hemostatically active forms. At sites of vascular injury, VWF exposed to subendothelial collagen acts as a molecular bridge, binding collagen via its A3 domain and platelet GPIb-IX-V receptor via its A1 domain, mediating platelet adhesion and thrombus formation under high shear stress. VWF also carries and stabilizes coagulation factor VIII in plasma, protecting it from premature proteolytic clearance. Deficiency or dysfunction causes von Willebrand disease. Subcellularly, VWF is stored in endothelial Weibel-Palade bodies and platelet alpha-granules, secreted into plasma, and deposited in the subendothelial extracellular matrix.
