Function
Tissue-type plasminogen activator (t-PA/tPA), encoded by the PLAT gene in humans, is the principal thrombolytic serine protease. It is synthesized and secreted primarily by vascular endothelial cells lining blood vessels and circulates in plasma. t-PA is a multi-domain glycoprotein (~70 kDa) containing a finger domain, growth-factor domain, two kringle domains, and a C-terminal serine-protease domain. It is largely inactive in free solution but is strongly activated upon binding fibrin within a thrombus, where it cleaves the zymogen plasminogen to generate plasmin, the enzyme that degrades fibrin. This fibrin-dependence localizes fibrinolysis to the clot and limits systemic proteolysis. Beyond hemostasis, t-PA participates in extracellular matrix remodeling, cell migration, and neuronal plasticity in the central nervous system. Recombinant t-PA (alteplase) is used clinically to dissolve occluding thrombi.
