Function
Chemokine (C-C motif) ligand 2 (CCL2), also called monocyte chemoattractant protein 1 (MCP-1) and, in the mouse, JE, encoded by the Ccl2 gene, is a pro-inflammatory CC-chemokine. It is secreted by mouse monocytes/macrophages, vascular endothelial cells, fibroblasts, astrocytes, and smooth muscle cells upon inflammatory stimulation. CCL2/JE signals through the G-protein-coupled receptor CCR2 on circulating monocytes, particularly the Ly6C-high inflammatory subset, as well as memory T cells and dendritic cells; CCR2-dependent signaling is required for the emigration of monocytes from the bone marrow into the blood and for their recruitment into inflamed tissues. The CCL2/CCR2 axis is the dominant pathway driving monocyte and macrophage recruitment in mouse models of peritonitis, atherosclerosis, myocardial infarction, neuroinflammation, and tumor growth, and Ccl2- and Ccr2-deficient mice show markedly impaired macrophage recruitment and host defense. CCL2 is a secreted chemokine immobilized on endothelial glycosaminoglycans to guide monocyte trafficking in vivo.
