Target info
Function
C-C motif chemokine ligand 5 (CCL5/RANTES), encoded by the Ccl5 gene in mice, is a pro-inflammatory CC chemokine orthologous to human CCL5. It is an approximately 8 kDa secreted protein produced by activated T lymphocytes, macrophages, platelets, fibroblasts, and endothelial cells. CCL5 is chemotactic for mouse monocytes, memory T cells, eosinophils, basophils, NK cells, and dendritic cells, recruiting these leukocytes to sites of infection and tissue injury. It signals through the G-protein-coupled receptors CCR1, CCR3, and CCR5 on target leukocytes, activating Gi-dependent phospholipase C, intracellular calcium mobilization, PI3K/Akt, and MAPK/ERK pathways that mediate integrin activation, cytoskeletal rearrangement, directed migration, and degranulation. It also engages the atypical decoy receptors ACKR1 and ACKR2 to shape and resolve chemokine gradients. In mouse models CCL5 contributes to antiviral and anti-tumor leukocyte recruitment, allograft rejection, allergic inflammation, and atherosclerotic plaque macrophage accumulation. CCL5 is a soluble secreted protein that associates with cell-surface heparan sulfate proteoglycans on endothelium to form tissue haptotactic gradients; it is not membrane-anchored and circulates in body fluids.
