Function
Growth/differentiation factor 15 (GDF15), originally identified as macrophage inhibitory cytokine-1 (MIC-1), is encoded in the mouse by the Gdf15 gene and is a divergent member of the transforming growth factor-beta (TGF-beta) superfamily. It is synthesized as a pro-protein that is furin-cleaved into a mature disulfide-linked dimer and secreted. Under basal conditions it circulates at low levels, but it is strongly induced in placenta and, upon cellular stress, injury, inflammation, or mitochondrial dysfunction, by many tissues including liver, kidney, heart, lung, and activated macrophages. Its only known high-affinity receptor is GFRAL (GDNF-family receptor alpha-like), a receptor whose expression is restricted to neurons of the area postrema and nucleus tractus solitarius in the hindbrain; GDF15 binds GFRAL together with its RET co-receptor, activating downstream AKT/ERK signaling that suppresses appetite and food intake, produces an aversive/nausea-like response, and regulates energy balance and body weight. Circulating GDF15 is a widely used biomarker of cellular stress, mitochondrial dysfunction, and disease severity. It is a secreted circulating cytokine.
