Function
Interleukin-13 (IL-13) is a Th2-type cytokine encoded in mice by the Il13 gene on chromosome 11. It is secreted as an ~13-kDa monomeric four-helix glycoprotein structurally related to IL-4. Il-13 is produced by activated CD4+ Th2 cells, group 2 innate lymphoid cells (ILC2s), mast cells, basophils, eosinophils, and nuocytes in response to IL-25 and IL-33 alarmins. It signals through the type II receptor composed of Il-13Ralpha1 (Il13ra1) paired with Il-4Ralpha (Il4ra); JAK1 and TYK2 are cross-activated and recruit STAT6 as the principal transcription factor. Phospho-STAT6 dimerizes, translocates to the nucleus, and drives GATA3-dependent type 2 effector genes. Il-13 also binds the high-affinity decoy receptor Il-13Ralpha2 (Il13ra2), which lacks a signaling-competent cytoplasmic tail and mediates ligand internalization and depletion of extracellular Il-13. Functionally, murine Il-13 stimulates B-cell proliferation and IgE class switching, induces goblet-cell mucus production and airway mucous metaplasia, promotes eosinophil recruitment via eotaxins, upregulates VCAM-1 on endothelium, drives airway smooth-muscle hyperreactivity, and induces fibroblast activation and collagen deposition in models of allergic asthma and pulmonary fibrosis. It also antagonizes Th1-driven pro-inflammatory cytokine synthesis partly through suppression of NF-kappa-B. The mature cytokine is secreted into the extracellular space and mucosal lumen; no membrane-bound isoform of Il-13 itself is described.
