Function
Interleukin-1 receptor antagonist (IL-1Ra; IL1RA), encoded by the mouse Il1rn gene, is a naturally occurring anti-inflammatory cytokine of the interleukin-1 family orthologous to human IL-1Ra. It is produced by monocytes, macrophages, epithelial cells, keratinocytes, and adipocytes and exists as a secreted protein released into the extracellular space and plasma, together with intracellular isoforms retained within producing cells. Mouse IL-1Ra binds the cell-surface signaling receptor IL-1R1 with high affinity comparable to IL-1alpha and IL-1beta but, unlike the agonists, does not recruit the accessory chain IL-1RAcP or trigger MyD88/IRAK-mediated NF-kappaB and MAPK signaling. It therefore acts as a competitive receptor antagonist that limits IL-1-mediated inflammation, fever, and autoinflammatory tissue damage; Il1rn-deficient mice develop exacerbated IL-1-driven inflammatory disease. It is predominantly a secreted/circulating cytokine, with intracellular variants confined to the cytosol and nucleus.
