Function
Interleukin-22 (IL-22), encoded by the mouse Il22 gene, is a secreted IL-10-family cytokine orthologous to human IL-22. It is produced by Th17/Th22 CD4+ T cells, gamma-delta T cells, NKT cells, neutrophils, and group 3 innate lymphoid cells at barrier sites, and is released into the extracellular space. Mouse IL-22 binds a heterodimeric receptor of IL-22R1 and the shared IL-10R2 chain; IL-22R1 is expressed on tissue cells (epithelial, stromal, and hepatocytes) but essentially absent from immune cells, restricting IL-22 action to non-hematopoietic targets. Receptor ligation activates JAK1 and Tyk2, which phosphorylate STAT3 (and STAT1/STAT5), along with PI3K/AKT and MAPK cascades, inducing epithelial cell survival, proliferation, and antimicrobial peptides including Reg3beta, Reg3gamma, S100A8/A9, and defensins. IL-22 thereby mediates mucosal barrier defense, wound healing, and protection against enteric and pulmonary pathogens, and is modulated by the soluble inhibitor IL-22BP. It is a secreted cytokine.
