Function
Interleukin-27 subunit p28 (IL-27p28; also termed IL-27 or IL-30), encoded by the mouse Il27 gene, is the alpha-like ligand subunit of the heterodimeric cytokine IL-27, orthologous to human IL-27p28. It is produced by activated dendritic cells and macrophages and noncovalently pairs with the separate Ebi3 gene product (EBV-induced gene 3) to form the secreted IL-27 heterodimer; free p28 can also act antagonistically. IL-27 signals through a receptor of IL-27Ralpha (WSX-1) plus gp130, activating JAK-STAT signaling dominated by STAT1 and STAT3. STAT1/T-bet drives early Th1 clonal expansion and synergizes with IL-12 for IFN-gamma, whereas STAT1-dependent and -independent branches suppress Th17 development and promote IL-10-producing Tr1 regulatory cells. IL-27 thus exerts both pro-Th1 and anti-inflammatory/immunoregulatory effects. It is a secreted cytokine; the p28 subunit itself is part of a secreted heterodimer.
