Function
Myeloperoxidase (MPO) is the mouse ortholog of human MPO, encoded by the Mpo gene on chromosome 11, a heme peroxidase of the peroxidase-cyclooxygenase superfamily. Like the human enzyme, murine MPO is synthesized during promyelocytic differentiation of the granulocyte lineage, cotranslationally N-glycosylated, processed by glucosidases in the ER and cis-Golgi, and finally proteolytically matured in prelysosomal/azurophilic granule compartments into a paired heavy-light chain heterotetramer. It is the most abundant protein packed into the azurophilic (primary) granules of mouse neutrophils and polymorphonuclear leukocytes, where it functions as a lysosomal luminal enzyme. When neutrophils phagocytose microorganisms, MPO is discharged into the phagolysosome, where it uses NADPH-oxidase-derived hydrogen peroxide and chloride to produce hypochlorous acid and related reactive oxidants that mediate oxygen-dependent microbial killing. During degranulation or NET formation, granule contents including MPO are also released into the extracellular space. Mouse Mpo mRNA is abundantly expressed in myeloid progenitor and neutrophil-like cell lines (e.g., NFS-60) and in mature circulating neutrophils and blood monocytes, but is largely absent from parenchymal cells such as brain microglia and normal neural tissue. Subcellular localization is lysosomal/azurophilic granular at rest and extracellular/secreted upon activation.
