Function
von Willebrand factor (VWF), encoded by the mouse Vwf gene, is a large multimeric plasma glycoprotein essential for hemostasis in mice. It is synthesized by vascular endothelial cells and megakaryocytes, stored in endothelial Weibel-Palade bodies and platelet alpha-granules, and secreted into the plasma and subendothelial matrix. VWF forms disulfide-linked multimers of varying sizes, with ultra-large multimers being the most adhesive. At sites of vessel injury, VWF binds exposed subendothelial collagen and platelet GPIb-IX-V receptors, bridging platelets to the damaged wall and mediating primary hemostasis under high shear. It also chaperones coagulation factor VIII in the circulation, stabilizing it and preventing its premature clearance. Vwf knockout mice recapitulate aspects of von Willebrand disease with prolonged bleeding. Subcellularly, VWF is stored in Weibel-Palade bodies and platelet alpha-granules and is secreted into plasma and the extracellular matrix.
